Webinar AFC SFM

The Microbiology-Cytometry SFM-AFC group offers a series of short Webinars to complement the “Microbiology” chapter of the book “Flow Cytometry” released by Lavoisier at the end of 2020. Each webinar will include the presentation of a method, detailing the protocols, for the study of microorganisms by flow cytometry, and a time for questions from participants.

Enumeration                              June 15  1:30 p.m. -2:15 p.m.

Flow cytometry allows rapid analysis and counting of microorganisms in a suspension. Associated with fluorescent markers of viability/mortality, it is possible to count in less than 30min the total, viable and dead yeasts or bacteria present in a suspension. This webinar will present methods and fluorescent markers for the enumeration of microorganisms, based in particular on the ISO 19344:2015 standard for the enumeration of lactic and probiotic bacteria in dairy ferments.

Flow-FISH                                         June 22  1:30 p.m. -2:15 p.m.

The FISH (Fluorescent In Situ Hybridization) method coupled with flow cytometry allows the specific and rapid detection of microorganisms in a mixture. After giving the principle, advantages and limits of this method, the commercial kit (VERMICON) for detecting Brettanomyces bruxellensis in wines will be presented by Pascal Brunou (SYSMEX)

Specific detection by antibody           January 14, 2022 1:30 p.m. -2:15 p.m.

Labeling a microorganism with a specific fluorescent antibody allows it to be detected quickly by flow cytometry. This webinar will present the principles of the method as well as elements on the choice of antibodies (polyclonal/monoclonal), the choice of fluorochrome and coupling (direct or sandwitch method), the possibility of simultaneous labeling with a viability marker……These data will be illustrated with examples.

Sorting of microorganisms              January 22, 2022 1:30 p.m. -2:15 p.m.

This webinar will present the principle, the sorting criteria (size, fluorescence) and the specificities to be taken into account when sorting microorganisms, in particular if the objective is to put them back in culture after sorting. In the case of strict anaerobic microorganisms, additional precautions are necessary to maintain anaerobiosis throughout the sorting operation. Different examples will be presented.
