IL-12 and IL-23 are closely related cytokines and represent the most proinflammatory mediators of the IL-12 superfamily. Both cytokines are structurally similar and share both producing and sensing cell types. While IL-12 is a canonical TH type I inducing mediator, IL-23 is firmly linked with type III immunity. Both cytokines are considered central mediators of TH cell polarization and are linked with inflammation and even immunopathology. However, whilst IL-12 stimulates type 1 lymphocytes and can eliminate established tumors, the loss of IL-12 leads to drastically enhanced inflammation across various models of chronic inflammatory disease and autoimmunity. Likewise, IL-23, which plays a non-redundant function in the development of autoimmunity and type 3 immune responses, it also increases tumor growth. These at first glance paradoxically appearing properties form the focus of this presentation. I will discuss the discoveries around IL-12 and IL-23 from a historical perspective, taking into account the most recent findings on their immunoregulatory properties in the context of inflammation and cancer in particular and immunity in general.

FSEV-AFC Hands-on Workshop 2024

The first HANDS-ON WORKSHOP: “Single Extracellular Vesicles analysis” is jointly organized by the French Society of Extracellular Vesicles (FSEV)and the French Association of Cytometry (AFC) and will take place from February 7 to 9, 2024 at FIAP Jean Monnet,, in Paris

Extracellular vesicles are gaining increasing attention from researchers and clinicians due to their pivotal roles in various physiological and pathological processes. They influence the progression and treatment responses of several diseases, including cancers. Additionally, extracellular vesicles hold promise as disease biomarkers, offering non-invasive diagnostic potential. The analysis of single extracellular vesicles is now emerging as the main challenge in the field of extracellular vesicles.

This workshop will cover the basic and advanced technology dedicated to single extracellular vesicles analysis and will consist of lectures by international experts in the field and hands-on pratical modules animated by the following companies: Abbelight, Agilent, BD, Beckman Coulter, Cytek Biosciences, Excilone & Particle Metrix, IZON, Miltenyi, Myriade and NanoFCM.

The workshop will be organized as follow over 3 days:

  • An afternoon dedicated to an overview of the fundamental and technological challenges in the study of extracellular vesicles by academic experts.
  • A full day with practical sessions divided into 4 slots of 2 hours where each company will allow groups of 6 people to manipulate directly their device and to test the reference material of the company and batches of calibrated vesicles.
  • A Morning dedicated to the analysis and the discussion of the results on each technology.

Abstract :
Triple negative breast cancer (TNBC) is the deadliest subtype of breast cancer Standard treatments for
patients with early stage, locally advanced or metastatic TNBC include the combination of neoadjuvant
chemotherapy and pembrolizumab, an immune checkpoint blockade targeting the programmed cell
death 1 (PD-1) receptor PD-1 blockade reinvigorate tumor antigen specific CD8+ T cells in primary TNBC.
However, approximately 40 and 80 of early stage and advanced or metastatic TNBC are resistant to
PD 1 blockade therapy, respectively.
Regulatory T cells (Tregs) are an immunosuppressive subset of CD4+ T cells essential for maintaining self
tolerance in physiological settings. Abundant tumor infiltration by Tregs harbouring hallmarks of highly
suppressive CD25high effector Tregs (eTregs) has been associated with resistance to PD 1 blockade in
cancer indications for which pembrolizumab was approved much earlier than for TNBC, such as non
small cell lung cancer. In contrast, in TNBC, the prognostic value of intratumoral Tregs remains
controversial and no association between baseline levels of eTregs and responses to PD 1 blockade
therapy has been reported, despite the clinical significance of this cancer indication. In the present
thesis, we used single cell technologies to dissect the nature and function of eTreg during TNBC
progression. We found that an immunosuppressive TNBC microenvironment resistant to PD-1 blockade
therapy is marked by an imbalance between effector CD8+ T cells and CD25 high eTregs. We further
identified CD25 as the most selective surface marker of eTregs in primary TNBC and associated
metastases for limited on target/off target effects. In a syngeneic TNBC model, selective Treg depletion
by a next generation anti-CD25 monoclonal antibody (mAb; ALD 2510; Alderaan Biotechnology)
synergized with PD-1 blockade to safely mediate durable tumor growth control by increasing effector
αβCD8+ T cell/Treg ratios in tumors and in the peripheral blood Overall, we propose that Treg depletion
by next generation anti-CD25 mAb could improve clinical responses to PD-1 blockade therapy in patients
with TNBC at different stage of the disease

EasyPanel Webinar: Automated/AI-guided Flow Cytometry Panel Designer

At the Cyto conference in Montreal 2 weeks ago, we picked up (flow-cytometry.net), a tool that uses AI to instantly designate optimized panels of cytometry (conventional and spectral). We therefore wish to communicate this information to the French cytometry community.

EasyPanel is a French startup that has developed an algorithm that automatically offers panels that minimize spillover/complexity/similarity score, match antigens to fluorochromes (based on expression/coexpression levels and fluorochrome brightness) and identify commercially available products (the tool is directly integrated with the commercial database of all suppliers, and is diagnostic of it).

About a hundred universities/pharma companies already use this tool, most of them in the US (but also a few in France: Pasteur, Servier, IGR…).

A webinar is therefore offered to you on June 29 to discover the interest of this software in your laboratories.

Hello, the French Cytometry Association (AFC) is organizing the Practical Days: Unsupervised analyzes in cytometry from Thursday July 6 to Friday July 7, 2023 in Sophia-Antipolis.

Programme des Journées Pratiques

Hosted by :

Goal :

  • Introduction to unsupervised analysis methods of multi-parametric data.
  • Presentation and use of algorithms offered by Tercen, FlowJo and Cytobank.
  • Algorithms presented: t-SNE, UMAP, FlowSOM…

Audience : Technicians, engineers, students, researchers regularly practicing cytometry.

Prerequisites :

  • Good knowledge in multiparametric cytometry
  • Knowledge of the R language is not a prerequisite
  • Mastery of good practices in cytometry.

Résumé : Le codage à barres CD45 des cellules vivantes implique la coloration d’échantillons de cellules immunitaires avec des combinaisons uniques d’anticorps anti-CD45 comme identificateurs d’échantillons. Cela permet le regroupement d’échantillons afin de les traiter comme un échantillon de cellule unique pour la coloration et l’acquisition des cellules. Il améliore le flux de travail, améliore la cohérence des données et réduit l’utilisation de réactifs.

Au cours de ce webinaire, nous discuterons de toutes les étapes d’une expérience de codage à barres CD45 sur cellules vivantes, de la conception du panel à l’analyse des données. Tout d’abord, lors du panel design, il est important de s’assurer que la coloration CD45 n’interfère pas avec la détection de marqueurs d’intérêt dans d’autres canaux. Ensuite, plusieurs ajustements doivent être apportés aux procédures de coloration et d’acquisition. Enfin, les fichiers FCS obtenus à partir d’échantillons de cellules à code-barres sont décodés pour attribuer chaque événement cellulaire à son échantillon d’origine.

La vitalité cellulaire dépend de la charge énergétique des cellules. Elle se traduit par la cinétique des réactions métaboliques ou activité métabolique. Dans les industries de fermentation, les ferments doivent être viables mais aussi actifs pour coloniser rapidement le milieu en début de fermentation. On peut aussi évaluer l’effet de stress sur les cellules par la mesure de vitalité. La cytométrie en flux permet de mesurer en 30min la vitalité cellulaire, ce qui est plus simple et plus rapide que les mesures cinétiques d’activités métaboliques. Ce webinaire présentera la méthode avec quelques exemples de corrélation avec des mesures d’activités sur des levures et des bactéries.

We are collecting information to assess the level of interest in educational materials in languages other than English for our “Lost in Translation: Cytometry in a Multilingual Setting” workshop at CYTO in Montreal which will take place on Sunday May 21 from 15:45 to 16:45 in room 511D.

Scientific communication at the higher level worldwide takes place in English and any scientist who wishes to advance their career internationally must be fluent. There is, however, a huge population of clinical, research and industrial cytometrists who are not English fluent and who are working at institutions where the primary language is not English. Often technical staff, both in research and clinical environments, are the ones operating the instruments and generating the data but are not able to follow training courses or teaching material in English. In this workshop we would like to address the issues encountered by English speaking or bilingual faculty teaching in countries where this has been a difficulty and explore ways to improve them. The second topic is to put into place a system to identify and gather educational material, whether in English or already existing in other languages, to translate into multiple languages and to make the translated material available for free access to all who can benefit.

Could you please diffuse this information and survey to anyone or groups who might be interested? Welcome any of the associated societies or persons who might be interested in participating in this endeavor for different languages to attend the workshop. 5ème Congrès FSEV 19-21 Octobre 2022

After two years of exchanging via screens, we are impatient to be able to bring together the community of EVologists during a face-to-face congress.

For the occasion, the FSEV (French Society of Extracellular Vesicles) is joining forces with the AFC (French Cytometry Association) to organize a congress in Paris from October 19 to 21, 2022.

This congress will offer you an afternoon dedicated to the contribution of flow cytometry technology in the study of extracellular vesicles.

Two days of plenary sessions, oral presentations and posters will then follow which will give you an overview of the vitality and excellence of French research on extracellular vesicles. We hope to see many of you there!

Registration and abstract submission are open. You can submit your abstracts until September 09, 2022. Registration is open until September 30, 2022.
